A membrane current underlying a sarcolemmal outward and inward rectifying current (I(out)/I(in)) in human erythrocytes was studied in the presence of internal and external concentrations of K+, NH4+, Rb+, and Cs+ to determine ion selectivity and permeation rate of the inward rectifier. The current was expressed as the difference between (a) a current with respect to an external K+ concentration, (b) the reversal potential of the current in relation to an internal K+ concentration, and (c) the reverse voltage for permeation of the current. A lumen-positive membrane potential was required for activation of the inward rectifier. Saturating the membrane with K+ was necessary for activation of the current, and K+ permeation rate was 5.5- to 10.0-fold greater than that of Na+ and 3.5- to 4.8-fold greater than that of Li+. K+ permeation rate of the inward rectifier is 1-10 times slower than that of Cl- and 10-15 times slower than that of Na+, but permeation of Rb+ was 300-fold faster than that of Na+ in the inward rectifier. External NH4+ or Rb+ could replace K+ and generate a current similar in magnitude to the current induced by K+. I(in) was inhibited in external NH4+ and Rb+ suggesting that the inward rectifier may be involved in NH4+ transport.
