The long-term goal of this work is to develop a safe, effective and inexpensive HIV vaccine. The HIV envelope glycoprotein, gp120/gp41 complex, consists of two non-covalently associated subunits that form a trimeric spike on the virion surface. Each subunit consists of a surface region, the gp120, and a transmembrane domain, the gp41. The gp120 is the major antigenic determinant for the induction of neutralizing antibodies. The immunogenic nature of gp120 makes it an ideal target for broadly neutralizing antibodies. In this project, we will develop novel monoclonal antibodies to the surface region of the gp120 glycoprotein and express them in a yeast expression system. The recombinant gp120 protein will be used as an immunogen to induce broadly neutralizing antibodies against HIV-1. We will also determine whether the broadly neutralizing antibody response is elicited by the viral envelope proteins of primary isolates or by the pseudoviruses.
